52. Abstract

Various procedures for preparing and modifying the alkali-resistant structural polysaccharide of the scales of the haptophycean alga, Pleurochrysis scherffelii, are described. Corresponding changes in the ultrastructural arrangement are revealed by electron microscopy and X-ray diffraction. In addition, the specific amino acid composition of a proteinaceous component associated with this structural polysaccharide is given. The data support the concept that cellulose is the predominant structural polysaccharide in these scales and appears to be firmly linked to polypeptide moieties with an extraordinarily high serine content. These compositional and structural results represent the final evidence that fibrils containing crystalline cellulose I are produced within the cisternae of the Golgi apparatus in this plant cell.
